“GTx-024 showed a dose-dependent improvement in total lean body mass and physical function and was well tolerated.”

Evidence level

Human randomized, double-blind, placebo-controlled phase 2 trial (120 healthy men over 60 and postmenopausal women).

Funding

Industry: all authors were "employees of and have stock and stock options with GTx, Inc."; GTx supplied the drug. Run at private UK/German clinical research units.

What was published

A 12-week trial of daily enobosarm (GTx-024) at 0.1 to 3 mg versus placebo.

What it shows

At 3 mg, participants gained "a statistically significant (P < 0.001) average of 1.3 kg" of lean mass versus placebo, with improved stair-climb power (P = 0.013). "No SAEs were reported."

What it does not show

Healthy volunteers over 12 weeks; it does not show benefit for bodybuilders or long-term safety. Products sold online as 'ostarine' are not the trial drug.

Documented counterpoints

HDL cholesterol fell 17% and 27% at 1 and 3 mg; total testosterone fell in men; ALT rose above normal in eight subjects, and one was withdrawn with ALT 4.2 times normal.
